Effects of Ethionine and Methionine on Acute Iron Intoxication

Abstract
Author stu3ied histopathologica¢¥: changes in various organs of rabbits induced acute iron into~-ication <<-ith 0.3 gm per kg orally and the effects of pretreatment with ethionine and/or methionine on the above changes. The ethionine (¢¥~00 mg;¢¥kg) and methionine (1 gm/kg) were devided into four doses and given intraperitoreaily even; the other day prior to the iron administration.
The results were as follows:
1) The most characteristic histopathologic lesions in the liver were zonal necrosis, fatty changeand parenchymal hemorrhage. These lesions were most severe in ethionine pretreated group, and the se~-erity was lessened in ethionine and methionine pretreated Group, control group and methionine pretreated group in order.
The necrotic changes, in ethionine, and ethionine ~tnd methionine pretreated groups, were prominent both in central and peripheral portion of lobules, whereas in control group the changes were marked in peripheral zone.
Such retrograde changes, in ethionine, and ethionine and methionine pretreated groups, were prominent both in central and peripheral portion of lobules, whereas in control group the changes were marked in peripheral zone.
Such retrograde changes of the liver were most severe in the animals of 6 to 24 hours after iron intoxication, but the changes were lessened in the animals of 2 days after intoxication, and quite minimal after a week.
The iron reaction of the liver tissues was mildly positive in animals of G to 24 hours of all experimental groups, but the reaction was not observed later on.
2) Major histopathologic lesions in the spleen were congestion and fibrosis. The congestion was tended to be marked in early experimental period while the fibrosis was in later period. And the iron reaction was moderately positive in early experimental period, but the degree of positivity was declined with time.
3) Histopathologic lesions in the gastrointestinal tract were congestion, edema and hemorrhagic necrosis of the gastric mucosa. Such findings were most severe in the animals of 6 to 24 hours. after iron intoxication, but the changes were lessened in the animals of 2 days after intoxication and not noted after a week.
The iron reaction in the mucosal surface also. showed strong positive in the animals of 6 to 24 hours after iron intoxication but negative in the animals after a week.
4) The testis showed mild atrophic changes in a few cases of control and ethionine pretreated groups.
5) The other organs such as lungs, pancreas,. kidneys, :slivery glands and adrenals showed acute congestion only in cases of 6 to 24 hours. after intoxication, and no other particular changes were noted.
